β-hydroxybutyrate impairs bovine oocyte maturation via pyruvate dehydrogenase (PDH) associated energy metabolism abnormality

Background: Ketosis is one of the most frequent and costly metabolic disorders in high-producing dairy cows, negatively associated with health reproductive performance bovine. mainly caused by accumulation ketone body β-hydroxybutyric acid its diagnosis based on β-hydroxybutyrate (βHB) concentration blood. Methods: In this study, we investigated effects βHB bovine oocyte maturation subclinical (1.2 mM) clinical (3.6 mM). Results: The results showed disrupted development capacity. Further analysis that induced oxidative stress mitochondrial dysfunction, as indicated increased level reactive oxygen species (ROS), structure distribution, depolarized membrane potential. Furthermore, triggered early apoptosis, shown enhanced levels Caspase-3 Annexin-V. Moreover, 3.6 mM disruption pyruvate dehydrogenase (PDH) activity, showing decrease global acetylation modification increase abnormal spindle rate. Conclusion: Our study subclinical/clinical had toxic function PDH which might affect energy metabolism epigenetic oocytes embryos.
